CARBOHYDRATE ANALYSIS
 

 

Analysis of monosaccharides and oligosaccharides is carried out using a Dionex DX 500 high pH anion exchange chromatography (HPAEC) apparatus. Primary monosaccharide content (and O- and N-linkages) of a glycoprotein can be ascertained using acid hydrolysis followed by HPAEC analysis. Both O- and N-linked oligosaccharides can be removed from glycoproteins by chemical (acid hydrolysis, hydrazinolysis) or enzymatic (endoglycosidases) means. Oligosaccharide structures can be elucidated by comparison of the chromatographic pattern with those of known standard structures. Further information on glycan structure can be obtained by lectin blotting, SDS-PAGE followed by PAS staining, and exoglycosidase digestion followed by HPAEC.

BITS is currently investigating the carbohydrate structures of recombinant antibodies in order to understand their role in producing functionally active protein. 

Carbohydrate analysis of purified glycoproteins can be undertaken on a contract basis.
